dc.description.abstractThe search for new drugs to treatment of tumor diseases has driven the study of substances with recognized biological activity, such as tetracycline analogues (TCs) known as CMTs (Chemically Modified Tetracyclines). The main characteristic is that they lack the dimethylamino group at the C4 position, responsible for the antibacterial activity, potentiating side effects such as enzymatic inhibition, more specifically, gelatinases (MMP-2 and MMP- 9), which are the component of the matrix metalloproteinase (MMPs) family. These enzymes can degrade the extracellular matrix allowing the migration of tumor cells by the body. Among the CMTs, CMT-3 is the one with the highest affinity for the catalytic domain of gelatinases. The mechanism of inhibition of MMPs by CMT3 is partially known, but studies using theoretical methodology have shown that the increased activity of CMT-3 is due to hydrophobic interactions with the S1 'pocket of the enzyme which is the main structural difference between gelatinases and remaining MMPs. In the present study seeking to increase the interaction, specificity, and consequently the inhibitory potential of CMT-3 against MMPs, 12 substituents at the C9 position of CMT-3 based on ligands derived from aminomethylcyclines and glycyclines were proposed, then these ligands were docked inside of the MMP-9 active site. After the molecular docking simulations, the two lower energy configurations for each complex were analyzed, with configuration 1, in which the ligand does not interact directly with the catalytic zinc, was the most stable. All ligands tested had greater binding energy than unmodified CMT-3, except for glycylcline derivatives. The CMT3-A21, CMT3-A1 and CMT3-A5 analogs obtained the highest binding energy to the enzyme. However, only the MMP-9:CMT3-A21 complex has a direct interaction with the catalytic zinc and the S1’ pocket of the enzyme, doing the binding energy of this complex 2.23 Kcal.mol-1 more stable than MMP-2:CMT-3 complex. Thus, the MMP-9:CMT-3A21 complex will be the focus of subsequent studies applying Molecular Dynamics (DM) simulations to elucidate the mechanism of enzyme inhibition by identifying specific regions of the ligands interaction with the biological receptor.pt_BR
dc.description.resumoA procura por novos fármacos para tratamento de doenças tumorais tem impulsionado o estudo de substâncias com reconhecida atividade biológica, como os análogos das tetraciclinas (TCs) conhecidos como CMTs (Chemically Modified Tetracyclines). A principal característica das CMTs é que essas não possuem o grupo dimetilamino na posição C4, responsável pela atividade antibacteriana, potencializando efeitos secundários como inibição enzimática, mais especificamente, das gelatinases (MMP-2 e MMP-9), componente da família das metaloproteinase de matriz (MMPs). Essas enzimas são capazes de degradar a matriz extracelular permitindo a migração de células tumorais pelo organismo. Dentre as CMTs, a CMT-3 é a que possui maior afinidade com o domínio catalítico das gelatinases. O mecanismo de inibição das MMPs pela CMT3 é parcialmente conhecido, porém estudos usando metodologia teóricas mostraram que a maior atividade da CMT-3 se deve a interações hidrofóbicas com o bolso S1’ da enzima o qual é a principal diferença estrutural entre as gelatinases e o restante das MMPs. No presente estudo buscando aumentar a interação, especificidade, e consequentemente o potencial inibidor da CMT-3 frente as MMPs, foram propostos 12 substituintes na posição C9 da CMT-3, baseados em ligantes derivados das aminometilciclinas e das glicilclinas, posteriormente esses ligantes foram ancorados dentro do sítio ativo da MMP-9. Após as simulações de docking molecular foram analisadas as duas configurações de menor energia para cada complexo, sendo a configuração 1, na qual o ligante não interage diretamente com o zinco catalítico a mais estável. Todos os ligantes testados obtiveram energia de ligação maiores do que a CMT-3 não modificada, a exceção dos derivados de glicilclinas. Os análogos CMT3-A21, CMT3-A1 e CMT3-A5 foram os que obtiveram maior energia de ligação com a enzima, porém apenas no complexo MMP- 9:CMT3-A21 ocorre interação direta com o zinco catalítico além da interação com bolso S1’ da enzima, fazendo com que a energia de ligação desse complexo fosse 2,23 Kcal.mol-1 mais estável que o complexo MMP-2:CMT-3. Sendo assim, o complexo MMP-9:CMT-3A21 será o foco dos estudos posteriores utilizando simulações de dinâmica molecular (DM) para elucidar o mecanismo de inibição da enzima identificando regiões específicas de interação do ligante com o receptor biológico.pt_BR
